Rapid and Efficient Regeneration of Populus ussuriensis Kom. from Root Explants through Direct De Novo Shoot Organogenesis

Populus ussuriensis is an important tree species with high economic and ecologic values. However, traditional sexual propagation time-consuming inefficient, challenging afforestation wood production using P. ussuriensis, requires a rapid efficient regeneration system. The present study established rapid, efficient, stable shoot method from root explants in several plant growth regulators. Most buds (15.2 per explant) were induced at efficiency under WPM medium supplemented 221.98 μM 6-BA, 147.61 IBA, 4.54 TDZ within two weeks. further multiplicated elongated 57.74 GA3 for four average number of elongation multiplication 75.2 78%, respectively. All the shoots rooted week none them showed abnormality rooting. time spent entire this direct organogenesis was seven weeks, much shorter than conventional indirect callus induction phase, no abnormal observed. This novel system will not only promote massive propagation, but also accelerate genetic engineering studies trait improvement species.
